МОЛЕКУЛЯРНО-ГЕНЕТИЧНІ МЕХАНІЗМИ ВИЖИВАННЯ І РЕЗИСТЕНТНОСТІ САЛЬМОНЕЛ

У статті наведено дані наукової літератури, що описують молекулярно-генетичні механізми захисту від несприятливих умов у бактерій роду Salmonella. Основними бар’єрами на початкових етапах інфекційного процесу є висока осмолярність та низькі значення pH. Захисні механізми сальмонели спрямовані на уникнення дії цих факторів з метою подальшої проліферації в інфікованому організмі.

Відповідь на несприятливі умови реалізується за допомогою двокомпонентних систем сигнальної трансдукції.
В основі яких лежить принцип взаємодії сенсора та регулятора. Сенсор, представлений гістидин-кіназою здійснює фосфорилювання регулятора, який у свою чергу прямо чи опосередковано ініціює транскрипцію генів патогенності. На сьогодні в науковій літературі описано функціонування системи EnvZ/OmpR, що активується за умов високої осмолярності та регулює утворення капсулярного Vi-антигену у тифоїдного серовара S. Typhi. Система PhoP/PhoQ здійснює відповідь на кисле середовище та функціонує за тим самим принипом: фосфорилювання регуляторного білка сенсором. Також охарактеризовано залежність системи PhoP/PhoQ від сигма-фактора RpoS (субодиниця бактеріальної РНК-полімерази). Сигналом для накопичення RpoS є низька концентрація катіонів Магнію.

З’ясновано, що дані системи сигнальної трансдукції регулюють транскрипцію оперонів, які кодують гени систем секреції білків 3 типу. До складу останніх входять ефекторні білки, здатні до перебудови цитоскелету епітеліоцитів тонкого кишечнику, що дозволяє сальмонелі проникати в них у вигляді специфічних фагосом.

Таким чином, двокомпонентні системи сигнальної трансдукції у сальмонели не лише є  механізмами, що забезпечують виживання та проліферацію в несприятливих умовах, але також здійснюють регуляцію генетичних детермінант, що кодують гени патогенності.
